The full name of the Tyrant was Tyrant T-001, also known as the Prototype Tyrant. This was what Umbrella Corporation intended to create as a super-soldier using the T-virus, but most attempts failed. The Tyrants currently wandering the base were not true Tyrants but semi-finished products, otherwise, they wouldn't be called prototypes.

Obtaining Tyrant's gene sample was simple: kill the Tyrant and extract its blood.

Could he just extract the blood?

Technically, yes. But extracting only blood was actually more troublesome than killing the Tyrant and then extracting blood. The Tyrant was no docile creature to allow its blood to be drawn willingly. In fact, any encounter with it would devolve into a furious battle until one side died.
